Application CommentsAnti-Bim recognizes an epitope that is common to all 3 Bim isoforms. Permeabilization of the cell membrane is necessary for Bim detection by immunofluorescence. Bim is a cytoplasmic protein present at low levels; to immunoprecipitate Bim, use 1-5 mg of total cell protein. Antibody should be titrated for optimal results in individual systems.

DescriptionPurified rat monoclonal antibody. Recognizes the ~18 kDa Bim, ~20 kDa BimL, and the ~24 kDa BimEL isoforms.
BackgroundBim, Bcl-2 interacting mediator of cell death, mRNA is differentially spliced to code for 3 isoforms of the Bim protein: Bim-110 amino acids, BimL-140 amino acids and BimEL-196 amino acids. The Bim proteins share a 9 amino acid BH3 motif with bcl-2 homologues and this BH3 region is required for bcl-2 binding. Bim proteins also bind to bcl-xL and bcl-w, but not to Adenovirus E1B19K and Epstein-Barr virus BHRF-1. In healthy cells, BimL binds to the cytoplasmic dynein light chain LC8 and is thereby associated with the microtubule associated dynein motor complex. An apoptotic stimulus that disrupts LC8 and the dynein motor complex allows Bim to interact with and neutralize bcl-2.
