Background InformationAlsin (UniProt: Q920R0; also known as Amyotrophic lateral sclerosis 2 protein homolog, ALS2) is encoded by the Als2 gene (Gene ID: 74018) in murine species. Alsin serves as as a GTPase regulator and controls survival and growth of spinal motoneurons. In adult mouse, it is predominantly expressed in the all regions of the brain and highest levels are found in the molecular layer of the cerebellum. During development, it is expressed by day E9.5, but its expression does not become predominant until early postnatal life. It is shown to be enriched in endosomal membrane fraction. Alsin is shown to contain a central Dbl-homology (DH; aa 684-879) and a pleckstrin-homology (PH; 895-1001) domain suggesting its guanine nucleotide exchange factor (GEF) activity for the Rho GTPases. At its C-terminal it contains a vacuolar protein sorting 9 (VPS9; aa 1507-1651) domain indicative of Rab5 GEF activity. Alsin also contains 5 regulator of chromatin condensation 1 (RCC1) repeats at its N-terminal side that indicate its GEF activity and its 8-membrane occupation and recognition nexus (MORN) repeats which are essential for its membrane attachment. Mutations in the Als2 gene are implicated in juvenile-onset amyotrophic lateral sclerosis and related conditions. (Ref.: Devon, RS et al., (2005). Neurobiol. Dis. 18(2); 243-257).
