Background InformationA disintegrin and metalloproteinase with thrombospondin motifs 1 (UniProt: Q9UHI8; also known as ADAM-TS 1, ADAM-TS1, ADAMTS-1, METH-1) is encoded by the ADAMTS1 (also known as KIAA1346, METH1) gene (Gene ID: 9510) in human. ADAM-TS1 is a zinc-binding protein that is synthesized with a signal peptide (aa 1-49) and a propeptide (aa 50-252) that are subsequently cleaved by furin endopeptidase to generate the mature form (aa 253-967). It has a conserved cysteine in the cysteine-switch motif that binds the catalytic zinc ion and inhibits the enzyme. The dissociation of cysteine from the zinc ion upon the activation-peptide release activates the enzyme. It also contains spacer domain (aa 725-849) and the TSO-1 domain that are important for a tight interaction with the extracellular matrix. ADAM-TS1 cleaves aggrecan, a cartilage proteoglycan at Glu1938-Leu1939 site within the chondroitin sulfate attachment domain and is involved in its turnover. ADAMTS1 is implicated in vascularization and development and displays anti- and pro-tumorigenic activities. ADAMTS1 knockout mice exhibit reduced body weight, kidney malformation, and impaired female fertility. (Ref.: Rodriguez-Baena, FJ., et al. (2018). Sci. Rep. 8: Article 13103).
