OverviewEMD Millipore’s MILLIPLEX® MAP Human Myokine Magnetic Bead Panel 15-plex kit to be used for the simultaneous quantification of any or all of the following analytes in human serum and plasma samples and in cell/tissue supernatants: Fractalkine/CX3CL1, BDNF, Oncostatin M (OSM), Osteonectin (SPARC), LIF, FGF-21, Erythropoietin (EPO), IL-15, IL-6, Apelin, Myostatin/GDF8, Irisin, Osteocrin/Musclin, FABP3 and Follistatin-Like Protein (FSTL 1).

MILLIPLEX® MAP offers the broadest selection of analytes across a wide range of disease states and species. Once the analytes of interest have been identified, you can rely on the quality that we build into each kit to produce results you can trust. In addition to the assay characteristics listed in the protocol, other performance criteria evaluated during the validation process include: cross-reactivity, dilution linearity, kit stability and sample behavior (e.g. detectability and stability). Background InformationMuscle contraction during physical activity is an important activator of the release of the myokines. Myokines not only can act in autocrine and/or paracrine manner in regulation of skeletal muscle growth, they can also act as endocrine hormones to mediate inter-organ crosstalk. The myokine theory provides molecular mechanistic explanations to the exercise-induced metabolic changes in liver and adipose tissue as well as the profound changes in immune and neuron systems.

Application NotesAlthough a two hour sample incubation may be used, for optimal sensitivity, use an overnight (16-18 hour) incubation. Serum and plasma samples require (recommend) a 1:2 dilution with Assay Buffer. Tissue culture supernatants may be run neat or appropriately diluted in control medium. Each well requires 25 μL of sample, standard or control.
