Description
OverviewAn antifungal antibiotic that acts as an inhibitor of nuclear export because of its ability to interact with and impair the function of the nuclear export factor CRM1. It is about twice as potent as Leptomycin A (Cat. No. 431051) in inhibiting nucleocytoplasmic translocation of Rev, blocks the Rev-dependent export of mRNA into cytoplasm, and suppresses HIV-1 replication in primary human monocytes (IC50 = 600 pM). In addition, LMB effectively induces the accumulation of p53 in the nucleus of normal fibroblasts as well as protects p53 from Mdm2-mediated ubiquitination and degradation.

DescriptionAn unsaturated, branched-chain fatty acid with a terminal lactone ring that acts as a potent inhibitor of CRM-1-mediated (exportin-1) nuclear export. Exhibits antibiotic, anti-fungal, and anti-tumor properties and blocks the progression of the cell cycle at the G1 and G2 phases. Shown to inhibit nucleo-cytoplasmic translocation of Rev at nanomolar concentrations and block Rev-dependent export of mRNA into the cytoplasm. Also shown to suppress HIV-1 replication in primary human monocytes (IC50 = 600 pM). Effectively induces the accumulation of p53 in the nucleus of normal fibroblasts and protects p53 from Mdm2-mediated ubiquitination and degradation. Useful for studies involving nuclear localization and protein trafficking in eukaryotic cells.
