OverviewBioTracker™ EP-HMRG is nearly nonfluorescent at pH 7 or higher. It fluoresces by the reaction with proteases that specifically digest the EP (glutamic acid, proline) peptide sequence. A green fluorescent product is membrane permeable and localizes to the lysosome.

SCT205 exhibits no or very low fluorescence at pH >= 7. It fluoresces by the reaction with proteases that digest the peptide sequces of EP (glutamic acid, proline).

SPECTRAL PROPERTIES:
Excitation: 500 nm
Emission: 524 nm
For fluorescence microscopy, blue excitation filter cubes for FITC or GFP are suitable.

Background InformationEarly detection of esophageal squamous cell carcinoma (ESCC) is an important for a favorable prognosis, but is difficult to achieve by conventional endoscopy. Since fluorescence-based visualization is considered a promising approach, development of an activatable fluorescence probe to visualize ESCCs became an urgent goal. Based on the fact that various aminopeptidase activities are elevated in cancer, specimens freshly resected from patients were screened with aminopeptidase-activatable fluorescence probes. The results indicated that dipeptidylpeptidase IV (DPP-IV) is specifically activated in ESCCs and would be an appropriate molecular target for detection of esophageal cancer. DPP-IV-targeted activatable fluorescence probes may prove useful tools for detection of cancers characterized by EP (glutamic acid-proline) sequences such as that found in DPP-IV (dipeptidyl peptidase.)

Application Notes
To use SCT205 EP-HMRG protease live cell probe, warm the vial to room temperature and add 30 μL of DMSO or PBS to one vial to make 1 mM solution.
Dilute the stock solution with a neutral buffer or culture medium to 1-2 uM and use the diluted solution immediately.

NOTE: Under acidic conditions, BioTracker™ EP-HMRG Green Protease Live Cell Probe emits fluorescence
by a different mechanism from protease hydrolysis. It therefore must be diluted with a neutral or weakly basic solution (pH>7).

Storage of the reconstituted probe solution is not recommended. If it becomes necessary to store temporarily, keep aqueous solution on ice, or freeze DMSO solution and use as quickly as possible.
