Background InformationDNA repair protein complementing XP-C cells (UniProt Q01831; also known as xeroderma pigmentosum group C-complementing protein, p125) is encoded by the XPC (also known as RAD4, XP3, XPCC) gene (Gene ID 7508) in human. Nucleotide excision repair (NER) removes damage to DNA caused by UV and mutagens. The NER process involves damage recognition and formation of an open DNA structure around the lesion, followed by dual incision, excision of an oligonucleotide of 24 to 32 residues, and gap-filling DNA synthesis to restore the DNA molecule. In human, the minimal set of components involved in NER includes replication protein A (RPA), XPA, XPC-RAD23B, XPG, ERCC1-XPF, TFIIH, replication factor C (RFC), PCNA, DNA polymerase δ or ɛ, and DNA ligase I. These factors can be divided into two groups, the six core factors necessary for damage recognition and dual incision (RPA, XPA, XPC-RAD23B complex, XPG, ERCC1-XPF, and TFIIH) and those for repair DNA synthesis in the gap-filling step. XPC is specifically involved in global genome NER (GG-NER) and serves as a damage recognition protein in complex with RAD23B. XPC is the rate-limiting factor in the GG-NER process, and XPC-dependent distortion recognition plays an essential role in initiating the recruitment of core NER proteins to the lesion.
