Background InformationVesicular glutamate transporter 2 (UniProt Q9JI12; also known as Differentiation-associated BNPI, Differentiation-associated Na(+)-dependent inorganic phosphate cotransporter, Solute carrier family 17 member 6, VGluT2) is encoded by the Slc17a6 (also known as Dnpi, Vglut2) gene (Gene ID: 84487) in rat. VGluT2 is a multi-pass membrane protein that mediates the uptake of glutamate into synaptic vesicles at presynaptic nerve terminals of excitatory neurons. Its N- and C-terminal tails (aa 1-71 and 499-582, respectively) are exposed at the cytoplasmic side. VGLUT1- and VGLUT2-expressing neurons represent the bulk of canonical glutamatergic neurons in the brain, with VGLUT1 representing the major subtype and accounting for approximately 80% of total vesicular transport of glutamate in the brain. Expression of VGLUT2 is reported to increase sharply during the first ten days of postnatal life, and also increases with increasing numbers of rhodopsin cells in the retina. It has been shown that both VGLUT1 and VGLUT2 are expressed prominently in the adult brain, however, during the first two weeks of postnatal development, VGLUT2 expression predominates. VGLUT1 expression becomes essential for survival at the time of the developmental switch from VGLUT2 to VGLUT1.
