Background InformationTriggering receptor expressed on myeloid cells 2 (UniProt: Q99NH8; also known as TREM-2, Triggering receptor expressed on monocytes 2) is encoded by the Trem2 (also known as Trem2a, Trem2b, Trem2c) gene (Gene ID: 83433) in murine species. TREM-2 is a single-pass type I monomeric membrane protein that forms a receptor signaling complex with TYROBP and triggers activation of the immune responses in macrophages and dendritic cells. TREM-2 is expressed at higher levels in the central nervous system (CNS), heart, and lung. At lower levels it is also expressed in lymph nodes and in non-lymphoid tissues such as kidney, liver and testis. In the CNS it is highly expressed in microglia, where it, along with its adaptor protein DAP12, regulates inflammatory cytokine release and phagocytosis of apoptotic neurons. It plays a role in chronic inflammation and may stimulate production of constitutive rather than inflammatory chemokines and cytokines. TREM-2 forms a receptor signaling complex with TYROBP and triggers activation of the immune responses in macrophages and dendritic cells. Apolipoprotein E (ApoE) is reported to function as a high-affinity ligand for TREM-2. ApoE binding is shown to enhance the phagocytosis of apoptotic N2a cells by primary microglia cells. Greatly reduced ApoE binding is seen with Alzheimer s disease-associated TREM2 R47H mutant. TREM2 is synthesized with a signal peptide (aa 1-18) that is cleaved off to generate the mature form with a large extracellular region (aa 19-171) that contains the V-type Ig-like domain, a single-transmembrane domain (aa 172-192), and a cytoplasmic tail (aa 193-227). The short cytoplasmic tail does not contain any signaling motifs and relies on its association with the immunoreceptor tyrosine-based activation motif- (ITAM-) containing TYROBP for signal transduction. Western Blotting Analysis: Cleaved soluble TREM2 (sTREM2) in the conditioned media of the cells. KO BV2 cells were created by CRISPR/CAS9 and were proven with commercial antibodies to be free of TREM2 expression. TREM2 is in fact highly N‐glycosylated and this may vary dependent on the cell line. Using Monocyte BV2 cells, because TREM2 is highly expressed in cells like macrophages and microglia. The mature band of TREM2 is 40‐60 kDa in the cell line BV2 with an immature precursor at around 36 kDa.
