Background InformationSrc Homology region 2 domain Phosphatase 1 (SHP-1) is also known as Protein Tyrosine Phosphatase 1C (PTP1C), Protein Tyrosine Phosphatase non-receptor type 6 (PTPN6), and Hematopoietic Cell Phosphatase (HCPH). SHP-1 is a SH2 domain-containing cytosolic tyrosine phosphatase characterized by a catalytic phosphatase domain of approximately 240 amino acids that selectively dephosphorylates tyrosine residues in proteins; the SH2 domains are critical for SHP-1 to bind to ITIM domains in inhibitory immunoreceptors. SHP-1 is highly expressed in hematopoietic tissues and has a negative impact on lymphocyte signaling. SHP-1 is implicated in the control of tyrosine kinase signaling pathways in cellular proliferation, with a potential role in cancer.

PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. N-terminal part of this PTP contains two tandem Src homolog (SH2) domains, which act as protein phospho-tyrosine binding domains, and mediate the interaction of this PTP with its substrates. This PTP is expressed primarily in hematopoietic cells, and functions as an important regulator of multiple signaling pathways in hematopoietic cells. This PTP has been shown to interact with, and dephosphorylate a wide spectrum of phospho-proteins involved in hematopoietic cell signaling.
