      Applications
ApplicationAnti-Myeloperoxidase Antibody, human white cell is an antibody against Myeloperoxidase for use in ELISA & WB.
      Key Applications
      • ELISA
      • Western Blotting
Application NotesImmunoblotting: antibody recognizes alpha and beta subunits of MPO. Reduced and denatured extracts of purified MPO produce blots produce 55-60kDa alpha subunits and 12-14kDa beta subunit bands140 Additional bands may be present due to cross reactivity with other myeloperoxidases.

      ELISA:

Optimal working dilutions must be determined by end user.

SpecificityMyeloperoxidase (Human White Cells). Monospecific by IEP. Some cross reactivity with Myeloperoxidase from other tissues and species may occur.

Myeloperoxidase (MPO) represents an early appearing and highly reliable intracellular myeloid lineage marker molecule. MPO protein can be detected in a subset of human hematopoietic bone marrow progenitor cells and in granulomonopoietic (GM) cells. However, other myeloid-related cell types such as epidermal Langerhans-type dendritic cells (LC) lack MPO. Therefore, human myeloid progenitors might be subdivided based on MPO protein expression into functional subsets.

      Entrez Gene Number
Entrez Gene SummaryMyeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of netrophils.
      Gene Symbol
      • MPO
      • myeloperoxidase
      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P05164 # Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity.
COFACTOR: Binds 1 calcium ion per heterodimer. & Binds 1 heme B (iron-protoporphyrin IX) group covalently per heterodimer.
SIZE: 745 amino acids; 83869 Da
SUBUNIT: Tetramer of two light chains and two heavy chains.
      SUBCELLULAR LOCATION: Lysosome.
DISEASE:SwissProt: P05164 # Defects in MPO are the cause of myeloperoxidase deficiency (MPD) [MIM:254600]. MPD is an autosomal recessive defect that results in disseminated candidiasis.
SIMILARITY: SwissProt: P05164 ## Belongs to the peroxidase family. XPO subfamily.
