      Background InformationMitogen-activated protein kinase 1 (UniProt: P63085; also known as EC:2.7.11.24; also known as MAP kinase 1, MAPK 1, ERT1, Extracellular signal-regulated kinase 2, ERK-2, MAP kinase isoform p42, p42-MAPK, Mitogen-activated protein kinase 2, MAP kinase 2) is encoded by the MAPK1 (also known as ERK2, PRKM1, PRKM2) gene (Gene ID: 26413) in murine species. ERK-2 is a widely expressed proline-directed serine/threonine protein kinase that is an essential component of the MAP kinase signal transduction pathway. Its protein kinase domain is localized to amino acids 23-311. ERK-2 binds both upstream activators and downstream substrates in multimolecular complexes. Its substrates include multiple transcription factors, cytoskeletal elements, regulators of apoptosis, and regulators of translation, and signaling molecules. MAPK/ERK cascade mediates diverse biological functions such as cell growth, adhesion, survival and differentiation through the regulation of transcription, translation, cytoskeletal rearrangements. ERK-2 is phosphorylated by MEK1 and MEK2 on threonine 183 and tyrosine 185 in the conserved TEY (Thr-Glu-Tyr) motif in response to external stimuli like insulin or NGF. Both phosphorylations are required for its activity. This phosphorylation causes conformational changes that enable full activation and interaction with its substrates.
