      Background InformationKinesin-like protein KIF1B (UniProt O60333; also known as Kinesin superfamily protein KIF1B, Klp) is encoded by the KIF1B (also known as KLP, CMT2, CMT2A, CMT2A1, HMSNII, KIAA0591, KIAA1448, NBLST1) gene (Gene ID 23095) in human. KIF1B is a member of the kinesin 3 family of proteins. The KIF1B gene encodes four alternatively spliced isoforms, among which isoform 2 and isoform 3 are also known as the alpha and beta isoforms. The alpha and beta isoforms share a common N-terminal motor domain but contain different C-terminal cargo domains. The alpha and beta isoforms are motor proteins implicated in anterograde transport of mitochondria and synaptic vesicle precursors, respectively. In addition, the beta isoform is required for developmental apoptosis induction by nerve growth factor (NGF) deprivation among neural crest progenitors during sympathetic neuron development. The beta isoform is reported to interact with RNA helicase A (DHX9), resulting in DHX9 nuclear accumulation and upregulated transcription of the proapoptotic XIAP-associated factor 1 (XAF1). KIF1B gene is located in a chromosomal region frequently deleted in neural crest–derived tumors, including neuroblastomas with loss of KIF1Bβ expression and impaired DHX9 nuclear localization.
