      Background InformationGlutamate receptor 1 (UniProt P42261; also known as GluR-1, AMPA-selective glutamate receptor 1, GluR-A, GluR-K1, Glutamate receptor ionotropic, AMPA 1, GluA1) is encoded by the GRIA1 (also known as GLUH1, GLUR1, GLURA) gene (Gene ID 2890) in human. Glutamate receptors are synaptic receptors located primarily on the membranes of neuronal cells. These receptors play important roles in neural communication, memory formation, learning, and regulation by mediating glutamate-dependent postsynaptic excitation. Glutamate receptors are divided into two types, ionotropic glutamate receptors (iGluRs) and metabotropic glutamate receptors (mGluRs). iGluRs form ion channel pores upon glutamate binding, while mGluRs indirectly activate ion channels on the plasma membrane via G protein-coupled signaling cascade. Although all iGluRs and mGluRs bind glutamate, they do exhibit different binding activities toward chemical ligands, such as NMDA, kainate, AMPA, L-AP4, ACPD, L-QA, and such distinciton is used to divide these receptors further into subtypes. Glutamate receptor 1 is an AMPA type iGluR, human GluR-1 is initially produced with an N-terminal signal peptide sequence (a.a. 1-18), the removal of which yields the mature multitransmembrane receptor.

      UniProt SummaryFUNCTION: Ionotropic glutamate receptor. L-glutamate acts as an excitatory neurotransmitter at many synapses in the central nervous system. Binding of the excitatory neurotransmitter L- glutamate induces a conformation change, leading to the opening of the cation channel, and thereby converts the chemical signal to an electrical impulse. The receptor then desensitizes rapidly and enters a transient inactive state, characterized by the presence of bound agonist.
