      • dedicator of cytokinesis 2
Background InformationDedicator of cytokinesis protein 2 (UniProt: Q8C3J5; also known as Protein Hch, Dock2) is encoded by the Dock2 gene (Gene ID: 94176) in murine species. Dock2 is a homodimeric protein that is predominantly expressed in immune cells and is involved in cytoskeletal rearrangements required for lymphocyte migration in response to chemokines. It is shown to be critical for migration, activation, proliferation, and effects of immune cells, including lymphocytes, neutrophils, macrophages, and dendritic cells. It is shown to activate RAC1 and RAC2, by functioning as a guanine nucleotide exchange factor (GEF) that exchanges bound GDP for free GTP. Dock2 plays an important role in the host immune response to SARS-CoV-2 infection and in the development of severe COVID-19. Its expression is suppressed in subjects with severe cases of COVID-19. UniProt SummaryFUNCTION: Involved in cytoskeletal rearrangements required for lymphocyte migration in response of chemokines. Activates RAC1 and RAC2 small GTPases, probably by functioning as a guanine nucleotide exchange factor (GEF), which exchanges bound GDP for free GTP. May participate in IL2 transcriptional activation via the activation of RAC2. Ref.1

SUBUNIT: Interacts with RAC1 and RAC2. Interacts with CRKL and VAV. Interacts with CD3Z By similarity.

SUBCELLULAR LOCATION: Intracytoplasmic membrane; Peripheral membrane proteinBy similarity. Cytoplasm › cytoskeletonBy similarity. Note= Colocalizes with F-actin By similarity.

TISSUE SPECIFICITY: Specifically expressed in hematopoietic cells. Ref.1

DOMAIN: The DHR-2 domain probably mediates the GEF activity By similarity.

SIMILARITY: Belongs to the DOCK family.

      Contains 1 DHR-1 (CZH-1) domain.

      Contains 1 DHR-2 (CZH-2) domain.
