      Key Applications
      • Immunohistochemistry (Paraffin)
Application NotesImmunohistochemistry on unfixed frozen sections and formalin-fixed paraffin embedded tissues. Antigen retrieval is recommended. HEIR or protease treatment can be used.

Western Blotting: AE1 recognizes LMW cytokeratins from the acidic subfamily including: Clone AE1 recognises the 56.5, 50, 50', 48 and 40kD human cytokeratins of the acidic subfamily.

Optimal working dilutions must be determined by end user.

SpecificityRecognizes most of the acidic (Type I) keratins. It is broadly reactive and stains positively almost all epithelia and their neoplasms. MAB1612 produces heterogeneous staining within a given epithelium; the detailed staining pattern varies depending on the differentiated and/or growth state of individual cells.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P05787 # Together with KRT19, helps to link the contractile apparatus to dystrophin at the costameres of striated muscle.
SIZE: 483 amino acids; 53704 Da
SUBUNIT: Heterotetramer of two type I and two type II keratins. keratin-8 associates with keratin-18. Associates with KRT20. Interacts with HCV core protein and PNN. When associated with KRT19, interacts with DMD. Interacts with TCHP.
      SUBCELLULAR LOCATION: Cytoplasm.
TISSUE SPECIFICITY: Observed in muscle fibers accumulating in the costameres of myoplasm at the sarcolemma membrane in structures that contain dystrophin and spectrin. Expressed in gingival mucosa and hard palate of the oral cavity.
PTM: Phosphorylation on serine residues is enhanced during EGF stimulation and mitosis. Ser-74 phosphorylation plays an important role in keratin filament reorganization.
      DISEASE:SwissProt: P05787 # Defects in KRT8 are a cause of cryptogenic cirrhosis [MIM:215600].
SIMILARITY: SwissProt: P05787 ## Belongs to the intermediate filament family.
MISCELLANEOUS: There are two types of cytoskeletal and microfibrillar keratin: I (acidic; 40-55 kDa) and II (neutral to basic; 56-70 kDa).
