Background InformationELL-associated factor 2 (UniProt: Q96CJ1; also known as Testosterone-regulated apoptosis inducer and tumor suppressor protein) is encoded by the EAF2 (also known as TRAITS, BM-040) gene (Gene ID: 55840) in human. EAF2 is an androgen induced tumor suppressor that is frequently down regulated in prostate cancer. It is ubiquitously expressed and is detected in heart, brain, placenta, skeletal muscle kidney, bone marrow, liver, and prostate. It acts as a transcriptional transactivator of ELL and ELL2 elongation activities. It serves as a potent inducer of apoptosis in prostatic and non-prostatic cell lines and is reported to inhibit prostate tumor growth in vivo. Overexpression of EAF2 in prostate cancer cell lines is shown to inhibit colony formation and xenograft tumor growth. EAF2 knockout mice are reported to develop late-onset lung adenocarcinoma, hepatocellular carcinoma, B-cell lymphoma, and high-grade prostatic intraepithelial neoplasia. Concurrent knockdown of EAF2 and p53 can induce alteration in several pathways associated with carcinogenesis and inactivation of EAF2 and p53 can activate STAT3 and drive prostate tumorigenesis.
