Background InformationCD226/DNAM1 is a T cell expressed plasma membrane receptor protein that plays multiple roles in lymphocyte signaling, intercellular adhesion, lymphokine secretion and cytotoxic T-cell and NK cell mediated killing. CD226 activates natural killer (NK) cell-mediated cytotoxicity and is a key player in the immune synapse between killer T cells and targets. Also, because it recognizes CD155 and nectins CD226/DNAM1 is being examined as a potential anti-cancer agent because soluble CD226 directly inhibits tumor growth by binding CD155 expressed on tumor cells and down regulating proliferation. CD226/DNAM-1 is expressed on Th1, but not Th2 or Th0 cells, it is also expressed on NK cells, CD8+ T-cells, but not developing T cells. CD226/DNAM-1 is also expressed on monocytes and platelets.
