Background InformationAutophagy-related 8a, isoform A and Autophagy-related 8b, isoform B (UniProt: Q9W2S2 and Q9VEG5, respectively) are encoded by the ATG8a and ATG8b genes (Gene ID: 32001 and 42132, respectively) in Drosophila melanogaster. Atg8 is a highly conserved and ubiquitously expressed protein that is upregulated under various stress conditions. Atg8 genes of multicellular animals is divided into three subfamilies LC3, GABARAP, GATE16 (depending on the animal). LC3 is the mammalian ortholog of Drosophila Atg8. Atg8 It shares a strong structural similarity to ubiquitin, despite the lack of similarity in amino acid sequence. The ubiquitin core of the Atg8 consists of a four-stranded central beta sheet, where the two central strands are parallel to each other and the two outer strands are antiparallel to the central strands. Between these strands are two alpha helices with alpha3 between beta2 and beta3 and alpha4 between beta3 and beta4. In resting cells, the majority of Atg8 is in found in an unlipidated state (Atg8-I) and during autophagy it is conjugated with a lipid moiety (Atg8-II). The lipidated form is the one that is recruited to autophagosomal membranes. The level of Atg8-II is an indicator of autophagosome activation. (Ref.: Shpilka, T., et al. (2011).
