      Overview

rLysozyme Solution, Veggie™ Grade is a special grade of rLysozyme prepared using certified animal-free or disease-free reagents. All of the steps in the preparation of the recombinant enzyme are carried out using reagents of nonanimal origin, with the exception of the IPTG used to induce protein expression. IPTG is chemically synthesized by a stringent process from D-galactose isolated from lactose, a milk sugar. The lactose is derived from certified disease-free cows. rLysozyme Solution, Veggie Grade has the same stability and specific activity as the rLysozyme Solution and requires no change to the protocol.

With its high specific activity (1700 KU/mg), only a small amount of rLysozyme Solution, Veggie Grade is required to break the cell wall of E. coli. Use the enzyme at 3-5 KU/g cells to enhance protein recovery from cells treated with BugBuster or BugBuster HT protein extraction reagents or with PopCulture Reagent. Or, treat thawed cell pellets with only the enzyme using 45-60 KU/g cells. Optimal protein purification is achieved by treating the cell lysate with Benzonase® Nuclease, which efficiently reduces extract viscosity by degrading the nucleic acids liberated as cells lyse. rLysozyme Solution, Veggie Grade is supplied as a ready-to-use solution at a concentration of 30 KU/µl in 50% glycerol containing 50 mM Tris-HCl, 0.1 M NaCl, 0.1 mM EDTA, 1 mM DTT, 0.1% TRITON X-100, pH 7.5. The solution is stable at -20°C.

rLysozyme Solution, Veggie Grade is a stabilized preparation of highly purified recombinant enzyme which has been prepared in the absence of animal-derived products. From the growth and induction of cells to protein purification and storage, all steps in the production of this enzyme use only certified animal-free reagents. Because of its high specific activity (1,700 KU/mg), only a small quantity of Veggie Grade rLysozyme is required to break the cell wall of E. coli. Use the enzyme at 3–5,000 units/g cells to enhance protein recovery from cells treated with BugBuster® Protein Extraction Reagent, BugBuster HT, or PopCulture® Reagents, or treat the cells with the enzyme alone at a ratio of 45–60 KU/g cells. The product is supplied as a ready-to-use solution at a concentration of 30 KU/microliter in 50% Glycerol containing 50 mM Tris-HCl (pH 7.5), 0.1 NaCl, 0.1 mM EDTA, 1 mM DTT, and 0.1% Triton X-100.
