      Background InformationLpin2, also known as Phosphatidate phosphatase LPIN2, and encoded by the gene Lpin2, plays important roles in controlling the metabolism of fatty acids. Lpin2 acts as a magnesium-dependent phosphatidate phosphatase enzyme which catalyzes the conversion of phosphatidic acid to diacylglycerol during triglyceride, phosphatidylcholine and phosphatidylethanolamine biosynthesis in the reticulum endoplasmic membrane. Lpin2 also acts as a nuclear transcriptional coactivator for PGC-1α protein. Lpin2 is localized to the cytoplasm and endoplasmic reticulum membrane but is also found in the nucleus. Lpin2 is expressed in many cells and tissues including liver, lung, kidney, placenta, prostate, and intestine. Expression is also seen in RBCs and at sites of lymphopoiesis in bone and spleen. Homozygous mutations in LPIN2 result in Majeed syndrome. Majeed syndrome is an autosomal recessive, autoinflammatory disorder characterized by chronic recurrent multifocal osteomyelitis and congenital dyserythropoietic anemia.
