      Background InformationProtein ENL (UniProt: Q03111; also known as YEATS domain-containing protein 1, MLLT1) is encoded by the MLLT1 (also known as ENL, LTG19, YEATS1) gene (Gene ID: 4298) in human. ENL is a component of the super elongation complex that is required to increase the catalytic rate of RNA polymerase II transcription by suppressing transient pausing by the polymerase at multiple sites along the DNA. It displays high homology with AF9/MLLT3 in its N-terminal YEATS domain and both are shown to be fusion partners of Mixed Lineage Leukemia (MLL), TrxG proteins. In the super elongation complex they connect it to other transcriptional factors and regulate transcriptional elongation. In response to double strand breaks both ENL and AF9 in the super elongation complex are phosphorylated by ATM, which increases the interaction between ENL in this complex and BMI1/RING1B in PRC1. The phosphorylated ENL recruits PRC1 at transcriptional elongation sites leading to the ubiquitination of histone H2A and results in transcriptional repression. Mutations in its YEATS domain have been linked to Wilms tumors. The mutant MLLT1 gene exhibits altered binding to acetylated histone tails and increased expression of Myc gene.
