      Applications
      Key Applications Immunoblotting (Western Blotting)
      Paraffin Sections
      Application NotesImmunoblotting (0.5-2 µg/ml)
      Paraffin Sections (see comments)
Application CommentsWill stain formalin-fixed, paraffin-embedded tissues, but requires titration for individual samples. The immunogen sequence is also conserved in rat caspase-1, but cross-reactivity has not been tested. Also recognizes cleaved forms of caspase-1 that retain amino acids 371-390. Antibody should be titrated for optimal results in individual systems.

DescriptionProtein G purified mouse monoclonal antibody. Recognizes the ~45 kDa latent form of caspase-1.
BackgroundCaspases are a family of cysteine proteases that play a key role in mediating apoptosis. All caspases are produced as inactive enzymes that require cleavage of a prodomain, followed by a second cleavage to generate the large and small subunits that form the tetrameric, active enzyme. Caspase-1, also known as ICE (IL-1β converting enzyme), was first isolated from monocytic lineage cells, and was so named because it converts the 33 kDa pro-form of IL-1β to the active, 17.5 kDa form via a specific, tightly controlled proteolytic cleavage process.
