Recombinant, human SIRT1 histone deacetylase consisting of amino acids 193-741 fused at the N-terminus to GST and expressed in E. coli. SIRT1 preferentially deacetylates histones H4 at K16 and H3 at K9 and interacts with and deacetylates histone H1 at K26. More than a dozen non-histone proteins have been found to serve as substrates for SIRT1. It plays a role in several biological processes including stress resistance, metabolism, differentiation, and aging. The active deacetylase is useful in the study of SIRT1 regulation and for inhibitor screening.
