      OverviewThe Periodic acid solution 0.5% for the PAS reaction(periodic acid Schiff) for the detection of aldehyde and mucosubstances in microscopy, is a ready to use solution used for the detection of mucopolysaccharides, glycogen, muco- and glycoproteins, glyco-, phospholipids, basal membranes and collagen. The material is treated with periodic acid creating aldehyde groups which react with Schiff's reagent and give a bright red color reaction, thus the nuclei appear blue.
      In the PAS reaction, the histological specimen material is first treated with periodic acid, resulting in the oxidation of the 1,2-glycols into aldehyde groups. The addition of Schiff’s reagent (fuchsin-sulfuric acid)(Cat.no. 109033) in the second step causes the aldehydes to react to form a brilliant red color. In the end result, the PAS reaction yields a specific color reaction with unsubstituted polysaccharides, neutral mucopolysaccharides, muco- and glycoproteins, and glyco- and phospholipids.
      The PAS reaction can be further combined with the Alcian blue staining method to detect mucosubstances (glycosaminoglycans).

      Acizii anorganici si anhidridele lor trebuie, mai Intai diluate sau hidrolizate prin amestecare cu grija in apa cu gheata, apoi neutralizate (manuşi de protectie, etuva) cu solutie de hidroxid de sodiu (Cod Merck 105587). Inainte de plasarea in container-ul D, verificati pH-ul cu strip-uri indicatoare de pH, universal (Cod Merck 109535). Acidul sulfuric fumans trebuie amestecat cu grija, picatura cu picatura, in acid sulfuric 40 %, (Cod Merck 109286) sub agitare. Asigurati-va ca aveti la indemana multa gheata penru racire. Cand s-a racit suficient, tratati acidul sulfuric foarte concentrat conform metodei descrise mai sus. Conform acestei proceduri, si alte anhidride pot fi convertite in acizii corespunzatori.Gazele acide (acid clorhidric, clor, fosgen, dioxid de sulf) pot fi barbotate in solutie diluata de hidroxid de sodiu si, dupa neutralizare, introduse in container-ul D.
