      BackgroundIn the developing nervous system, axon guidance and the establishment of appropriate neuronal connections involves a number of chemoattractant and chemorepellent molecules. Netrins are related to the extracellular matrix protein laminin and comprise a family of secreted molecules capable of guiding the growth cone. The first netrin family member to be characterized, UNC-6 from C. elegans, was initially identified from the gene mutation which lead to defects in circumferential axon migration in the nematode. In contrast, the identification of the first vertebrate netrins, designated netrin-1 and netrin-2, resulted from a biochemical rather than genetic approach. Netrin homologs have subsequently been identified from a number of species. Results from several studies suggest a role for netrin-1 in mediating chemotropic and outgrowth-promoting activities of floor plate cells to guide commissural axons towards the ventral midline. DCC (Deleted in Colorectal Cancer) is believed to function as a component of a receptor complex which mediates the effects of netrin-1 on commissural axons and helps coordinate the navigation of axons to establish precise patterns of neural connectivity. In addition to the netrins and their receptors, a number of other diffusable or membrane-associated ligand families and their receptors have been implicated in axon guidance, including the semaphorins, ephrins, and cell adhesion molecules.
