      Background InformationTumor necrosis factor receptor superfamily member 5 (UniProt: P27512; also known as B-cell surface antigen CD40, Bp50, CD40L receptor, CD40) is encoded by the Cd40 (also known as Tnfrsf5) gene (Gene ID: 21939) in murine species. CD40 is a single-pass type I transmembrane glycoprotein that serves as a receptor for TNFSF5/CD40L. It is a costimulatory protein found on antigen-presenting cells and is required for their activation. Its expression has been observed in B cells, basal epithelial cells, macrophages, follicular dendritic cells, endothelial cells, and a subset of CD34+ hematopoietic progenitors. It transduces TRAF-6 and MAP3K8-mediated signals that activate ERK in macrophages and B cells that leads to induction of immunoglobulin secretion. CD40 is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 20-193), a transmembrane domain (aa 194-205), and a cytoplasmic domain (aa 216-289). Five isoforms of CD40 have been described that are produced by alternative splicing.
