DescriptionNovel analogs capable of exhibiting Ins(1,4,5)P3-like agonist or antagonist effects on calcium release are more resistant to hydrolysis and metabolic degradation than IP3 or IP4. Consequently, they can produce a prolonged activation of cellular processes dependent on the Ins(1,4,5)P3 receptor interaction. These tools are important in the elucidation of the molecular interactions of Ins(1,4,5)P3 with its intracellular receptor and with Ins(1,4,5)P3-kinase and 5-phosphatase. Acetoxymethyl esters of inositol phosphate are highly lipophilic. Hence, they easily diffuse through the cell membrane. Note: Each set contains 1 µmol of Bt3(1,3,5)IP3/AM and 10 mg of PLURONIC® F-127 detergent (Cat. No. 540025) to aid in the solubilization of this product.

Recommended reaction conditions
Protocol for Treating Cells with IPx 1. Add 10 µl DMSO to 1 µmol Bt3(1,3,5)IP3/AM to make a 100 mM stock solution. 2. Prepare a 10% PLURONIC® F-127 solution in DMSO. DO NOT FREEZE! 3. Obtain 1 ml uniform cell suspension and place 500 µl of the suspension in a test tube. 4. Add 1 µl of the DMSO/Bt3(1,3,5)IP3/AM solution (step 1) and 2 µl of the 10% PLURONIC®/DMSO stock solution (step 2) to the cell suspension in step 3 and mix. 5. Add the cells/Bt3(1,3,5)IP3/AM /PLURONIC® mixture from step 4 to a tube containing the remaining 500 µl cell suspension. Mix carefully. The total test-tube volume now equals 1 ml.
