Background InformationAfadin isoform 1 (UniProt: O35889; also known as Afadin adherens junction formation factor, Protein Af-6) is encoded by the Afdn (also known as Af6, Mllt4) gene (Gene ID: 26955) in rat. Afadin is a multi-domain F-actin-binding homodimeric protein that belongs to an adhesion system. Adherens junctions are important in cell-cell contacts linking cells into a continuous sheet. Afadin plays a role in the organization of homotypic, interneuronal and heterotypic cell-cell adheren junctions. It interacts with cell adhesion molecules, cytoskeletal components, and signaling molecules and is generally considered to function as an adaptor protein. Two isoforms of Afadin have been described. Isoform 1 (I-Afadin) is widely expressed, including in heart, brain, spleen, lung, liver, skeletal muscle, kidney and testis. Isoform 2 (s-Afadin) is mainly expressed in the brain. Isoform 1 is reported to increase the viscosity of F-actin in a dose-dependent manner. Isoform 1 causes F-actin to associate into bundles and meshworks. Isoform 1 has one PDZ domain at the middle region, three proline-rich domains at the region following the PDZ domain, and one-Factin-binding domain at the C-terminal region.
