Background InformationSonic hedgehog protein (UniProt Q15465; also known as HHG-1, SHH) is encoded by the SHH (also known as HPE3, MCOPCB5, TPTPS, SMMCI) gene (Gene ID 6469) in human. Sonic Hh belongs to the hedgehog protein family, members of which also include Indian Hh and Desert Hh. Hh family members are involved in the cell fate and patterning during embryonic development, homeostasis, and adult tissue renewal. Similar to other family members, SHH binds to the patched (PTC) cell surface receptor, releasing the signal transducer Smoothened (Smo) to transduce the Hh signal further intracellularly, leading eventually to target genes transcription activation. The C-terminal domain of the precursor SHH displays an autoproteolysis activity and a cholesterol transferase activity, causing a cleavage of the precursor SHH into an N-terminal fragment (N-product; a.a. 24-197) and a C-terminal fragment (C-product; a.a. 198-462), as well as the covalent attachment of a cholesterol moiety to the C-terminal end of the N-product. The N-product is the active signaling species, whereas the C-product has no signaling activity. In addition, N-palmitoylation of N-product Cys24 by the palmitoylacyltransferase Hhat is required for N-product multimerization and full activity. SHH mutations and polymorphisms are linked to various developmental defects, including HPE3 (holoprosencephaly type 3), LSS (Laurin-Sandrow syndrome), MCOPCB5 (Microphthalmia, isolated, with coloboma, 5), SMMCI (Solitary median maxillary central incisor), and TPTPS (Triphalangeal thumb-polysyndactyly syndrome).

Immunohistochemistry Analysis: A representative lot detected SHH immunoreactivity localized to the outer root sheath of hair follicles in smoothened agonist SAG- (Cat. No. 566660) treated skin samples from wild-type mice, while SHH immunoreactivity was detected in the dermal cysts in SAG-treated skin samples from Vdr−/− mice (Lisse, T.S., et al. (2014). Mol.
