Background InformationMyocardin-related transcription factor A (UniProt: Q969V6; also known as MRTF-A, MKL/myocardin-like protein 1, Megakaryoblastic leukemia 1 protein, Megakaryocytic acute leukemia protein) is encoded by the MRTFA (also known as KIAA1438, MAL, MLK1) gene (Gene ID: 57591) in human. MRTF-A is a ubiquitously expressed transcription coactivator that associates with serum response factor (SRF) transcription factor to control expression of genes that regulate cytoskeleton during development, morphogenesis, and cell migration. The MRTFA-SRF complex activity responds to Rho GTPase-induced changes in cellular G-actin concentration, and thereby couples cytoskeletal gene expression to cytoskeletal dynamics. It is reported to bind to G-actin via its RPEL repeats. It is detected in cytoplasm as well as in the nucleus and its subcellular location is tightly regulated by actin. High levels of G-actin in the nucleus, observed during serum deprivation, result in low levels of nuclear MRTF-A, while reduced levels of nuclear G-actin result in accumulation of MRTF-A in the nucleus. G-actin-binding in the cytoplasm inhibits its nuclear import by masking the nuclear localization signal (NLS). In contrast, binding to nuclear G-actin promotes its nuclear export to the cytoplasm. Phosphorylation of serine 6 by ERK inhibits the binding of G-actin, unmasking the NLS and promoting nuclear import. Its N-terminal region is required for its nuclear localization and the C-terminal region mediates its transcriptional activity. (Ref.: Weissbach, J., et al. (2016). Mol. Cell.
