DescriptionImmunoaffinity purified rabbit polyclonal antibody. Recognizes the ~100 kDa Klotho protein.
BackgroundInactivation mutation of the recently discovered klotho gene in mice causes a syndrome resembling aging. Homozygous mutant klotho (kl/kl) mice exhibit premature aging phenotypes such as osteoporosis, age related skin changes, ectopic calcification, atrophy of genital organs and thymus, atherosclerosis, emphysema, ataxia, and short life span. The kl gene encodes a membrane protein whose extracellular domain carries homology to β-glucosidases. There also exists a splice variant of kl mRNA that encodes a putative secreted protein in both human and mouse. kl gene expression was observed to be tissue specific. Improvement of systemic aging phenotypes in kl/kl mice occurs even when the exogenous expression was limited to some organs, suggesting that KL associated aging is regulated through a humoral signaling pathway.
