Background InformationClaudin-5 (UniProt: O00501; also known as Transmembrane protein deleted in VCFS, TMDVCF) is encoded by the CLDN5 (also known as AWAL, TMVCF) gene (Gene ID: 7122) in human. Claudin-5 is a multi-pass membrane protein with three cytoplasmic domains (aa 1-7; 103-122; and 181-218); four transmembrane domains (aa 8-28, 82-102, 123-143, and 160-180); and two extracellular loops (aa 29-81 and 144-159). It plays a major role in tight junction-specific obliteration of the intercellular space. It is localized specifically to the endothelial cell layer in the brain, lung, liver, kidney, and skin. In peripheral tissues, it displays greater heterogeneity in tissue expression. At the blood-brain barrier, it is one of the most enriched tight junction proteins and its dysfunction has been linked to several neurodegenerative diseases, including Alzheimer s disease and multiple sclerosis.
