SpecificityIn Immunoprecipitation and Western blot experiments the Ki-S1 antibody recognizes a major protein of 170 kD which was identified as the a isoform of topoisomerase II (Boege et al., 1995; Kreipe et al., 1993). In addition, it has been shown that the Ki-S1 antibody recognizes a carboxyterminal a-isoenzyme specific epitope missing in topoisomerase IIb (Boege et al., 1995). In immunohistochemistry the Ki-S1 antibody shows strong nuclear staining only in proliferating cells. The epitope recognized by the antibody is also detectable in paraffin-embedded tissue sections (Kreipe et al., 1993). Accordingly, it has been shown that the expression of topoisomerase IIa is strongly restricted to proliferating cells (Tsutsui et al., 1993). The topoisomerase IIa antigen is preferentially expressed during G 1 , S, G 2 and M phase of the cell cycle, while resting, non-cycling cells (G 0 phase) lack topoisomerase IIa. In addition, constantly proliferating cells (e.g. cell lines) react positively to topoisomerase IIa during the entire cell-cycle. This specificity of Ki-S1 antibody for proliferating cells might make it a useful tool for determination of the proliferative fraction in solid tumors such as mammary carcinomas (Kreipe et al., 1993; Sampson et al., 1992; Camplejohn et al., 1993; Kreipe et al., 1993; Rasbridge et al., 1994) and gangliomas (Wolf et al., 1994).

Entrez Gene SummaryThis gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This nuclear enzyme is involved in processes such as chromosome condensation, chromatid separation, and the relief of torsional stress that occurs during DNA transcription and replication. It catalyzes the transient breaking and rejoining of two strands of duplex DNA which allows the strands to pass through one another, thus altering the topology of DNA. Two forms of this enzyme exist as likely products of a gene duplication event. The gene encoding this form, alpha, is localized to chromsome 17 and the beta gene is localized to chromosome 3. The gene encoding this enzyme functions as the target for several anticancer agents and a variety of mutations in this gene have been associated with the development of drug resistance. Reduced activity of this enzyme may also play a role in ataxia-telangiectasia.
