Background InformationMetabotropic glutamate receptor 5 or mGluR5 (UniProt Q3UVX5; also known as G protein-coupled receptor GRM5, G protein coupled receptor family C, group 1, member E) is encoded by the Grm5 (also known as Gprc1e, Mglur5, AI850523, 6430542K11Rik) gene in murine species (Gene ID 108071). The metabotropic glutamate receptors belong to a family of G protein-coupled receptors that have been divided into 3 groups based on their sequence homology, putative signal transduction mechanisms, and pharmacologic properties. mGluR1 and mGluR5 constitute Group I metabotropic glutamate receptors. mGluR5 is found on postsynaptic terminals of neurons and microglia. mGluR5 signals via Gq/G11 coupling to activate phospholipase C, resulting in calcium mobilization and activation of protein kinase C (PKC). In addition, mGluR5 is also reported to mediate signaling involving Src-family protein tyrosine kinase in a G protein-independent manner.

Glutamatergic neurotransmission is involved in most aspects of normal brain function and can be perturbed in many neuropathologic conditions. The metabotropic glutamate receptors are a family of G protein-coupled receptors, that have been divided into 3 groups on the basis of sequence homology, putative signal transduction mechanisms, and pharmacologic properties. Group I includes GRM1 and GRM5 and these receptors have been shown to activate phospholipase C. Group II includes GRM2 and GRM3 while Group III includes GRM4, GRM6, GRM7 and GRM8. Group II and III receptors are linked to the inhibition of the cyclic AMP cascade but differ in their agonist selectivities. Alternative splice variants of GRM8 have been described but their full-length nature has not been determined.
