Customized HPLC Columns & Applications

HPLC Columns

Merck offers a wide range of very successful and innovative packing materials for HPLC, manufactured in the unique and easy-to-use LiChroCART® cartridge system with its manu-CART® holder. In addition to proven and successful packing materials such as LiChrosorb®, LiChrospher® and Superspher® (irregular, regular and perfectly spherical silica gel sorbents), special sorbents such as Aluspher® (Al2O3) and several chiral phases such as the highly pure Purospher® and Purospher® STAR stationary phases are also available, and they ensure highest performance and efficient chromatography today and tomorrow.

LiChrosorb® – A Successful Packing Material From the Start!

Merck's LiChrosorb® is one of the most successful and reliable packing materials used in HPLC. LiChrosorb® packing materials offer the complete program of non-polar derivatives (RP-8, RP-18, RP-select B), polar derivatives (Si 60 and Si 100) and medium-polar derivatives (NH2, CN and DIOL). In addition to the analytical cartridges and columns such as LiChroCART® 250-4 and Hibar® RT 250-4, Merck offers LiChroCART® 250-10 semi-preparative cartridges as well as Hibar® RT 250-10 ready-to-use columns, packed with different LiChrosorb® packing materials.

LiChrospher® – Spherical Silica Carrier for Consistently High Results

LiChrospher® packing materials are reliable and versatile spherical silica carriers that are available in various modifications. The LiChrospher® CN, LiChrospher® NH2 and LiChrospher® DIOL polar-modified phases and the non-modified LiChrospher® Si are best for normal phase HPLC.

LiChrospher® PAH is a highly efficient and selective HPLC column especially designed for high-resolution separation of PAH; similarly, LiChrospher® WP 300 RP-18 is a highly selective and reliable HPLC column for the separation of peptides and low-molecular-weight proteins.

Furthermore, LiChrospher® packing materials are available as Hibar® RT ready-to-use columns and as LiChroCART® cartridges in different lengths and different internal diameters (10 mm, 4 mm, 3 mm and 2 mm). LiChroCART® 3-mm ID and 2-mm ID narrow-bore cartridges for HPLC save costs by reducing the use of solvents and allow the handling of very small quantities while increasing sensitivity and resolution.

Superspher® – First Choice for Highly Efficient HPLC

For highly efficient HPLC of complex mixtures where high peak capacity is required, Merck offers you Superspher®. These high-performance spherical silica carriers with a mean particle size of 4 µm yield the best pressure/separation performance ratio of today’s HPLC systems, according to both theoretical calculation and practical experience.

A broad range of modifications to Superspher® are available: non-polar derivatives (RP-8, RP-8 endcapped, RP-18, RP-18 endcapped and RP-select B) and polar derivatives (Si 60). In addition, Superspher® packing materials are available as LiChroCART® cartridges in different lengths and different internal diameters (4 mm, 3 mm and 2 mm).

Advanced formulation technology enables the production of spherical alumina particles as a base for Aluspher® 100 Al and Aluspher® 100 RP select B. Aluspher® 100 RP-select B is ideal for the use of basic eluents, as it suppresses ionization of basic compounds and avoids peak tailing. It is an alkali-stable reversed-phase stationary phase designed for the separation of neutral, basic and acidic compounds when basic eluents are used. Due to its stability in the pH 2-12 range, Aluspher® 100 RP-select B allows the use of basic eluents such as NaOH.

Purospher® HPLC columns are based on a high-purity, metal-free silica for excellent separations with very good peak symmetry. Due to the absence of heavy metals in the silica matrix and to the complete coverage of the silica surface, this stationary phase enables peak-tailing-free chromatography of acidic, basic and chelating compounds. This is of particular advantage for method development.

Purospher® HPLC Macking Materials

  • Purospher® RP-18 enables the peak-tailing-free elution of basic and chelating compounds, as well as the separation of hydrophilic compounds with a high percentage of water in the mobile phase.
  • Purospher® RP-18 endcapped is designed both for the separation of basic compounds with simple neutral eluents and for the elution of strongly acidic compounds.
  • Purospher® STAR RP-18/RP-8 endcapped is excellently suited for the separation of acidic, basic and chelating compounds with increased detection sensitivity. The high stability up to pH 10.5 allows the separation of highly basic compounds with alkaline eluents.
