Application CommentsDoes not cross-react with other Smad proteins. For immunoblotting incubate membrane with antibody diluted with TBS containing 5% BSA, 0.1% Tween®-20 detergent overnight at 4°C with gentle shaking. Please note that this is a rabbit monoclonal antibody and requires the use of a secondary antibody. Antibody should be titrated for optimal results in individual sytems.

DescriptionPurified rabbit monoclonal antibody generated by immunizing rabbits with the specified immunogen and fusing splenocytes with an immortal cell line. Recognizes the ~60 kDa Smad1/5 protein phosphorylated at Ser463/465.
BackgroundBone morphogenetic proteins (BMPs) constitute a large family of signaling molecules that regulate a wide range of critical processes including morphogenesis, cell-fate determination, proliferation, differentiation, and apoptosis. BMP receptors are members of the TGF-β family of Ser/Thr kinase receptors. Ligand binding induces multimerization, autophosphorylation, and activation of these receptors. Subsequently, the activated receptor phosphorylate Smad1 at Ser463 and Ser465 in the C-terminal motif, SSXS, as well as Smad5 and Smad8 at their corresponding sites. These phosphorylated Smads dimerize with Smad4 and translocate to the nucleus, where the transcription of target genes is stimulated.
