Application CommentsIt has not yet been determined which specific histone H1 phosphorylation sites this antibody detects. Does not cross-react with unphosphorylated histone H1. Also detects bands at ~64 kDa and ~130 kDa that have been shown to contain histone H1 using anti-histone H1 blotting. For immunocytochemistry, colcemid-treated HeLa cells were fixed with 2% paraformaldehyde followed by cold methanol. Variables associated with assay conditions will dictate the optimal working dilution.

DescriptionProtein A purified rabbit polyclonal antibody. Recognizes the ~32 kDa phosphorylated form of histone H1 protein.
BackgroundHistone H1, the nonnucleosome linker histone, becomes phosphorylated at multiple sites (3-6 sites/molecule of H1) upon entry of the cell into mitosis, most likely by Cdc2 kinase. It has been proposed that H1 phosphorylation may allow for transient chromatin decondensations, thereby allowing access of specific DNA binding factors involved in gene activation, DNA replication and chromosome condensation to the DNA.
