DescriptionThe mitochondiral permeability transition pore (PTP) is a proteinaceous aqueous pore of about 3 nm diameter that allows compounds of less than 1500 daltons to pass through. High levels of cytosolic Ca2+ and excessive concentrations of reactive oxygen species are known to contribute to the opening of the PTP, which depolarizes the mitochondria. The opening of PTP can be blocked by a decrease of pH, increase of ADP and Mg2+ concentrations. The PTP participates in the regulation of matrix Ca2+ levels, pH, and the volume. The PTP consists of an adenine nucleotide translocator (ANT) on the inner mitochondrial membrane and a voltage-dependent anion channel (VDAC) on the outer mitochondrial membrane. Hence, the PTP appears to be mainly localized in contact sites between the two membranes. The ANT is a specific antiporter that accounts for the exchange of ATP and ADP on inner membrane. VDAC, on the other hand, serves as the primary permeability pathway for metabolite flux between cytoplasmic and mitochondrial compartments. In the low-conductance state, the opening of PTP is pH dependent, which permits the diffusion of only small ions and then closes spontaneously. However, in the high-conductance state, the channel is stabilized in an open position and allows water and bigger molecules to enter the protein rich matrix. The mechanism of PTP formation and its biological significance have important implications in cellular bioenergetics, Ca2+ signaling, and apoptosis.
