Recommended reaction conditions
Protocol for the Assay of Hirudin Reagents: 1. 100 NIH units/ml thrombin 2. 9 mg/ml isotonic saline (0.9%) 3. 0.5% fibrinogen in isotonic saline 4. hirudin reconstituted with 1.0 ml of distilled H2O, isotonic saline, or other suitable buffer Assay: 1. Add 10-100 µl of the sample to be assayed to 200 µl of 0.5% fibrinogen solution and mix well. 2. Add 5 µl aliquots of a standardized thrombin solution (100 NIH units/ml) at one min intervals with mixing at 20°C. 3. The end-point is reached when a fibrin clot appears within one minute. Calculate the number of antithrombin units (ATU) of hirudin present in the solution. Note: This protocol is meant to serve only as a guideline. It may need to be modified for specific applications.
