A collection of antibodies for studying the Bcl-2 family of proteins. Contains 20 µg of each of the following antibodies: Anti-Bcl-2 (Cat. Nos. OP60 and PC68), Anti-Bax (Cat. No. PC66), Anti-Bcl-x (Cat. Nos. PC67 and PC89), and Anti-Bak (Cat. Nos. AM03 and AM04).

Application CommentsBcl-x (Ab-1) will react strongly with Bcl-xL and moderately with Bcl-xs; Bcl-xs (Ab-1) will not react to Bcl-xL. Brain sections can be used as a negative control for Bcl-2 and Bcl-xL (only random cells will stain positive); trp E (Ab-1) or normal rabbit IgG can also be used. MCF-7 cells treated with 0.2 mg/ml doxorubicin can be used as a positive control for Bcl-x (treat for 72 h) and Bax (treat for 48 h). The number and intensity of extra bands can be minimized by using 0.3% Tween®-20 detergent in the antibody and wash buffers. Antibodies should be titrated for optimal results in individual systems.

DescriptionSet of 7 antibodies specific for various members of the Bcl-2, Bax, Blc-x, Bcl-xs, and BAK.
BackgroundThe Bcl-2 gene family is critical for regulation of apoptosis and contributes to the pathogenesis of many diseases. Constitutive expression of Bcl-2 in some lymphomas and leukemias causes escape of these cells from physiological apoptosis in the germinal centers and appears to be a major element of pathogenesis. Members of the Bcl-2 gene family can act as either effectors or inhibitors of apoptosis. Among the proteins which promote or induce apoptosis are such proteins as Bcl-xs, Bax, and Bak. The inhibitors of apoptosis include Bcl-2 itself and Bcl-xL. Members of the Bcl-2 family of proteins form homodimers as well as form heterodimers with other family members and it is the intracellular balance between these various dimers which appears to determine whether a cell enters into the apoptotic pathway.
