Background InformationTransient receptor potential A1 (also known as Transient receptor potential ankyrin 1) is an excitatory ion channel on primary sensory nerves of the pain pathway. It is encoded by the TRPA1/ANKTM1 gene in human (Trpa1/Anktm1 in rat & mouse) and plays a critical role in pain associated with tissue injury, inflammation, and nerve lesions. TRPA1 is reported to colocalize with PAR2 and coexpress with TRPV1 (Transient receptor potential vanilloid 1) in rat dorsal root ganglion (DRG) neurons. Agonist-induced TRPA1 current is shown to be enhanced upon PAR2 cleavage/activation by proinflammatory trypsin & tryptase (PMID 17571167). TRPA1 activation, in turn, is reported to result in enhanced TRPV1 phosphorylation and activity in a calcium- and PKA-dependent manner (PMID 24275229).
