BackgroundPlk1 (ACC# NP_005021) is an evolutionarily conserved member of the polo-like serine/threonine kinase family that plays a critical role in spindle formation, chromosome segregation, regulation of the anaphase-promoting complex (APC), and execution of cytokinesis. Predominantly cytoplasmic during interphase, Plk1 is associated with condensed chromatin during mitosis when its kinase activity peaks. Plk1 regulates mitotic events through phosphorylation of its substrates Cdc25, the activator of Cdc2, and Cdc27, a component of the APC. Phosphorylation of Cdc25C by Plk1 is thought to lead to nuclear accumulation of Cdc25 and activation of Cdc2. Phosphorylation of Cdc27 leads to destruction of the APC. Ectopic expression of Plk1 in NIH3T3 leads to transformation. Over-expression of Plk1 is positively correlated with aggressiveness and prognosis of many cancers and inhibition of Plk1 activity results in growth arrest and apoptosis in cancer cells. Plk1 has been shown to be elevated in numerous cancers including lung, breast, ovarian, melanoma, colorectal, and gliomas.
