Background InformationN-methyl-D-aspartate receptor subunit NR1 (NMDAR1) is a multi-pass membrane protein belonging to the glutamate-gated ion channel family and is one of two subunits comprising the tetrameric protein NMDAR localized throughout the brain at excitatory synapses. NMDAR1 is mediated by glycine and is a critical component to excitotoxicity, learning, memory acquisition, synaptic plasticity, and synaptogenesis. It also plays a regulatory role for neuronal functions involving glutamate neurotransmission and the targeting of NMDA receptors on the cell surface. As a glutamate-gated ion channel, NMDAR1 is highly calcium permeable and possesses sensitivity to magnesium in a voltage-dependent manner.
