Background InformationLaminin subunit b1 (UniProt: P07942; also known as Laminin B1 chain, Laminin-1 subunit beta) is encoded by the LAMB1 gene (Gene ID: 3912) in human. Laminins are the major non-collagenous constituent of basement membranes that participate in wide variety of biological processes, including cell adhesion, differentiation, migration, signaling, neurite outgrowth and metastasis. They are a family of heterotrimeric extracellular glycoproteins consisting of α, β, and γ chains that are bound to each other by disulfide bonds into a cross-shaped molecule comprising one long and three short arms with globules at each end. Laminins mediate the attachment, migration, and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. Thus far five α, three β, and three γ subunits are described that assemble into at least 15 different laminin molecules. Different α, β, and γ chain isomers combine to give rise to different heterotrimeric laminin isoforms, which are designated by Arabic numerals in the order of their discovery. Laminin- β1 is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to generate the mature form. It is expressed in most tissues that produce basement membranes and is one of the 3 chains constituting laminin 1. It is shown to be involved in the organization of the laminar architecture of cerebral cortex and is probably required for the integrity of the basement membrane/glia limitans that serves as an anchor point for the end feet of radial glial cells and as a physical barrier to migrating neurons.
