Background InformationImmunity-related GTPase family M protein 1 (UniProt: Interferon-inducible GTPase 3, Interferon-inducible protein 1, LPS-stimulated RAW 264.7 macrophage protein 47, LRG-47, IRGM1) is encoded by the Irgm1 (also known as Ifi1, Iigp3, Irgm) gene (Gene ID: 15944) in murine species. IRGM1 belongs to the TRAFAC class dynamin-like GTPase superfamily that is expressed in lung and in primary macrophages. It is an interferon-inducible Golgi membrane associated GTPase whose disruption is shown to enhance susceptibility to various pathogens in mice. It behaves more like an integral membrane protein that is recruited to the plasma membrane forming phagocytic cups and remains associated with maturing phagosomes. Its association with phagosomes depends on the on nucleotide-binding but is independent of interferon-gamma. In mice it is required for interferon-gamma mediated clearance of acute protozoan and bacterial infections. Its deficiency has been linked with a severe immunodeficiency syndrome. It is shown to regulate host resistance to acute Toxoplasma gondii infections through their ability to inhibit parasite growth within the macrophage. Mice deficient in IRGM1 are shown to be severely leukopenic when infected with mycobacteria. Two isoforms of IRGM1 have been described that are produced by alternative splicing. Both isoforms localize to the Golgi apparatus and weakly to mitochondria, however, only the long isoform is detected in endolysosomal membranes. Clone 1B2 detects only the long isoform.
