Background InformationGamma-aminobutyric acid receptor subunit 1 (UniProt: P14867; also known as GABA(A) receptor subunit alpha-1) is encoded by the GABRA1 gene (Gene ID: 2554) in human. GABA is the major inhibitory neurotransmitter in the central nervous system that inhibits the generation of the action potential of the neuron. It plays an important role in the formation of functional inhibitory GABAergic synapses in addition to mediating synaptic inhibition as a GABA-gated ion channel. It is involved in the pathogenesis of certain neurological and psychiatric disorders. GABA interacts with the GABAA and GABAB receptors, which are widely distributed throughout the nervous system and in a variety of cell types. On binding of two GABA molecules to the extracellular receptor domain, a conformational change is induced in the oligomer and Cl moves into the cell leading to an inhibitory hyperpolarization. GABAA and GABAB receptors differ in their pharmacological, electrophysiological, and biochemical properties. GABA 1 receptor subunit is part of the heteropentamer formed by a combination of 1, β2, and 2 subunits. GABA receptor 1 subunit is synthesized with a signal peptide (aa 1-29) which is subsequently cleaved off to generate the mature form that contains four transmembrane domains (aa 252-273; 279-300; 313-334; and 422-443), one extracellular domain (aa 28-251), and a cytoplasmic domain (aa 335-421).
