BackgroundEPLIN (Epithelial Protein Lost in Neoplasm, Accesion No. NP 057441) was originally identified as the product of a gene that is down-regulated in prostate and breast cancer cell lines. In humans, EPLIN is expressed as two isoforms, EPLIN-α (~67 kDa) and EPLIN-β (~85 kDa), as a result of alternative promoter usage from a single gene on chromosome 12. The EPLIN-β isoform differs only from the EPLIN-α isoform by an additional 160 amino acids at the N-terminus. EPLIN-α expression is preferentially down-regulated in transformed cells. EPLIN contains a LIM domain that may facilitate dimerization and protein association. Expression of EPLIN increases the assembly of cellular stress fibers and inhibits membrane ruffling induced by active Rac. EPLIN stabilizes preformed filaments through its ability to interact with and bundle actin filaments. Loss of EPLIN may contribute to transformation due to rapid turnover of filaments associated with motility.
