Background InformationMembrane cofactor protein (UniProt: P15529; also known as TLX, Trophoblast leukocyte common antigen, CD46) is encoded by the CD46 (also known as MCP, MIC10) gene (Gene ID: 4179) in human. CD46 is a single-pass type I membrane glycoprotein that acts as a cofactor for complement factor I, a serine protease that protects autologous cells against complement-mediated injury by cleaving C3b and C4b deposited on host tissue. It is expressed in all nucleated cells and is synthesized with a signal peptide (aa 1-34), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 35-343), a transmembrane domain (aa 344-366), and a cytoplasmic domain (aa 367-392). The cytoplasmic domain has two tails that are termed as Cyt1 and Cyt2. CD46 also contains four Sushi domains that are required for its interaction with various human viruses. Sushi domains 1 and 2 are required for interaction with human adenovirus B PIV/FIBER protein and with Measles virus H protein; Sushi domains 2 and 3 are essential for Herpesvirus 6 binding and domain 3 is required for Neisseria binding. Sushi domains 3 and 4 are required for interaction with Streptococcus pyogenes M protein and are considered to be the most important for interaction with C3b and C4b. CD46 exhibits both signaling and receptor properties in immune and nonimmune cells, most of which are dependent on the expression of its cytoplasmic tails. Activation of CD46 is reported to promote T cell activation and differentiation and co-stimulation of TCR with CD46 can lead to T cell proliferation. Activated CD46 also leads to Tr1 Treg differentiation and secretion of IL-10 and granzyme B. The Tr1 differentiation initiated by CD46 is attributed to its Cyt1 domain.
