Background InformationBone marrow stromal antigen 2 (UniProt Q8R2Q8; also known as Bst-2, CD317, DAMP-1 protein homolog, HM1.24 antigen, Tetherin) is encoded by the Bst2 (also known as 2310015I10Rik, C87040, GREG) gene (Gene ID 69550) in murine species. Bst-2 is a type II transmembrane glycoprotein that exists as a disulfide bond-linked homodimer on the membrane. In addition, Bst-2 is reported to shuttle between the cell surface and the Golgi apparatus. Bst-2 is expressed in type I IFN-producing cells (IPCs), also known as plasmacytoid dendritic cells (DCs), which function as early responders to viral infections. Bst-2 is also known as tetherin because it prevents the release of enveloped viruses from the host cells by tethering the virions in infected cells. Murine Bst-2 is initially produced with a C-terminal propeptide (a.a. 153-172) sequence, the removal of which yields the mature protein with a large extracellular region (a.a. 52-152), a transmembrane segment (a.a. 31-51), and a short cytoplasmic tail (a.a. 1-30).

Key Applications
  • Flow Cytometry
  • Western Blotting
  • Immunocytochemistry
  • Inhibition
Application NotesWestern Blotting Analysis: A representative lot detected Bst2 in 50 µg mouse spleen tissue lysate (Courtesy of Dr. Youhai Chen, University of Pennsylvania School of Medicine, USA).
Immunocytochemistry Analysis: A representative lot detected Bst2 immunoreactivity among murine stomal OP9 cells and human embryonic kidney HEK293T cells transformed to express type I INF using retroviral vector MiGr1 (Courtesy of Dr. Youhai Chen, University of Pennsylvania School of Medicine, USA).
Flow Cytometry Analysis: A representative lot detected a high- and a low-Bst2-expressing populations among murine mesenteric lymph node (LN) cells (Courtesy of Dr. Youhai Chen, University of Pennsylvania School of Medicine, USA).
Inhibition Analysis: A representative lot, when administered via tail vein injection (200 µg/mouse), protected mice from death upon Listeria monocytogenes infection (Courtesy of Dr. Youhai Chen, University of Pennsylvania School of Medicine, USA).
